Abstract

The current research provides evidence to determine the rutin in the Melia azedarach plant by the use of chromatographic and spectroscopic analysis. Thin layer chromatography (TLC), UV-Visible spectroscopy, Fourier-transform infrared spectroscopy (FTIR) and high-performance liquid chromatography (HPLC) were used to compare the plant extract with standard rutin. The TLC analysis revealed that the standard and the extract had a similar Rf value; hence, they exhibited similar chromatographic characteristics. The UV-visible spectrum showed typical absorption bands of 200-220 nm and 255-270 nm, which indicated the presence of flavonoids in the compound. FTIR analysis showed the presence of functional groups including hydroxyl (OH), carbonyl (C=O), aromatic (C=C), and ether functional groups (C-O-C), which are also in line with the structure of rutin. HPLC analysis revealed that the plant extract had a significant peak at 2.912 min, which was like the retention time of standard rutin (2.803 min). In the present study, rutin was isolated, purified and characterized from the leaves of Melia azedarach by using chromato-graphic and spectroscopic methods. The findings suggest that the plant can be a source of the possible natural rutin that has antioxidant, anti-inflammatory and cardioprotective effects. Quantitative analysis and evaluation of biological activities are suggested to be carried out in further studies.
